Mesulid is usually used for an average short-term treatment in the case of people who are suffering from any of the following medical conditions: painful extra-articular medical disorders; acute pain, the symptomatic treatment of painful osteoarthritis, symptomatic Osteoarthritis, post-operative dental inflammation and pain, primary dysmenorrheal, etc.

Nimesulide, a preferential COX-2 inhibitor, has a multifactorial mechanism of action that affects the activity of MMPs (metalloprotease) and other biochemical markers of joint destruction, reduces the release of ROS (reactive oxygen species) and other toxic substances from neutrophils, and reduces the production of proinflammatory cytokines. Nimesulide has a rapid onset of the analgesic action.

These unique characteristics make nimesulide an appealing therapeutic choice in the treatment of acute pain. The therapeutic effects of Nimesulide are the result of its specific mode of action which targets a number of key mediators of the inflammatory process such as: COX-2 mediated prostaglandins, free radicals, proteolytic enzymes and histamine.

Mesulid is a non-steroidal anti-inflammatory drug (NSAID) with analgesic and antipyretic properties and a specific mode of action. Nimesulide is a sulphonanilide analogue; it is not related to conventional NSAIDs, which usually present a carboxyl or hydroxyl functional group. Mesulid is proved safe and effective in the treatment of a wide range of inflammatory and painful conditions, including osteoarthritis, extra-articular disorders such as tendinitis and bursitis, post-operative pain, primary dysmenorrhoea.

In addition, nimesulide acts as a free radical scavenger and interferes with the production of oxygen radicals by leukocytes without inhibiting leukocyte chemotaxis and phagocytosis. Since the products of free radicals directly and indirectly damage the host tissue and contribute to the maintenance of the inflammatory process, nimesulide helps protect against the tissue damage that occurs during inflammation and short-circuits the inflammatory process. In this manner, nimesulide exerts a potent anti-inflammatory effect in vivo even greater than that of its anti-prostaglandin effect.

Mesulid rapidly and effectively provides relief of pain and the signs and symptoms of inflammation associated with a wide array of disorders. Mesulid has especially proved useful in users who do not respond adequately to other NSAIDs, and in patients who are NSAID-intolerant due to hypersensitivity (e.g., asthmatics) or gastric intolerance.

Directions for use

The recommended daily dosage for adults and children over 14 is 100 mg twice daily, with plenty of fluid and preferably after meals. Mesulid is indicated for short-term therapy. The duration of treatment depends on the specific condition being treated but is usually between 3-14 days. For patients with osteoarthritis, short cycles of therapy for symptomatic relief are recommended.

Side effects

Mesulid safety profile is in line with that of the whole NSAID class. Mesulid shows a superior gastrointestinal safety profile in comparison to other NSAIDs and a low propensity for renal, cardiovascular, skin and hepatic side effects. Some side effects may occur: nausea, epigastric discomfort, abdominal cramps, heartburn, diarrhea, vomiting, etc.

During your treatment with Mesulid, you should avoid taking Hepatotoxic/Nimesulide Drugs (this also includes Augmentin, Other NSAIDs), hepatotoxins (for example anti-convulsants, anti-fungals, anti-tuberculous medicines like isoniazid, tacrine, amiodarone, pemoline, methyldopa, methotrexate, etc). If you have to start taking any of the medicines that have been listed here, you might not be allowed to continue your treatment with Mesulid, or you may have to take in a lower dose of this medicine. The drug is contraindicated in children below 12 years.
